Heart attacks damage cardiac muscle by cutting off oxygen and nutrients, and dead cells are replaced by scar tissue that cannot contract. A new biodegradable patch reported in Cell Biomaterials uses a microneedle array to deliver interleukin-4 (IL-4) directly to the injured heart surface. Huang describes the patch as a bridge: the microneedles penetrate the outer layer to reach the muscle beneath.
Each microneedle contains microscopic particles loaded with IL-4. When placed on the heart, the needles dissolve and release IL-4 into damaged tissue. IL-4 shifts macrophages from a pro-inflammatory state to a reparative, healing state. This immunomodulation limits scar formation and supports recovery of heart function, which could improve patient outcomes.
Researchers also observed changes in cardiomyocytes: the muscle cells became more communicative and more responsive to signals from nearby endothelial cells, which line blood vessels. The patch reduced inflammatory signaling from endothelial cells and increased signaling through the NPR1 pathway, a route that supports blood-vessel health and helps heart function.
At present the patch requires open-chest surgery for placement. Huang hopes to develop a version deliverable through a small tube to make the treatment less invasive and more practical for clinical use. The team plans to optimize patch design and delivery methods. Huang is collaborating with Xiaoqing (Jade) Wang to build an AI model mapping immune responses.
